Detection principle

The kit uses a two-antibody two-step sandwich enzyme-linked immunosorbent assay (ELISA). Add standard and sample to be tested to pre-coated CC Chemokine Receptor 5 (CCR5) transparent enzyme label plate. After incubation for a sufficient time, unbound components are washed away. Then, substrate A and B are added sequentially. The TMB substrate is converted to a blue product under horseradish peroxidase (HRP) catalysis, which turns yellow when acid is added. The color intensity is directly proportional to the concentration of CC Chemokine Receptor 5 (CCR5) in the sample. The OD value is measured at 450 nm, and the sample concentration is calculated based on the standard curve.

Steps】

1. Preparation: Take the kit out of the refrigerator and let it reach room temperature for 30 minutes.

2. Dilute the 20x concentrated washing solution with distilled water to the working concentration.

3. Add standard and sample: Place the enzyme-labeled plate on the frame. Set up standard wells, sample wells, and blank control wells. Add 50 μL of standard to each standard well. Add 10 μL of sample and 40 μL of diluent to each sample well (diluted 5 times). Do not add anything to the blank control.

4. Incubate at 37°C for 30 minutes.

5. Wash the plate: Discard the liquid, blot dry with absorbent paper. Fill each well with washing solution, let stand for 1 minute, discard the liquid, and repeat 4 times.

6. Add enzyme working solution: Add 50 μL to each well. Do not add to the blank control.

7. Incubate again for 30 minutes.

8. Wash the plate again following step 5.

9. Add color reagent A and B: Add 50 μL of each to each well. Incubate in the dark at 37°C for 15 minutes.

10. Stop the reaction: Add 50 μL of stop solution to each well. The color changes from blue to yellow.

11. Measure: Read the blank well first, then measure all wells within 15 minutes at 450 nm.

12. Calculate: Use the standard curve to calculate the sample concentration based on its OD value. Multiply by the dilution factor to get the actual concentration.

Sample requirements

1. Samples must not contain sodium azide (NaN₃), as it inhibits horseradish peroxidase (HRP).

2. Samples should be processed immediately after collection. If not tested right away, store at -20°C and avoid repeated freezing and thawing.

3. Ensure samples are fully centrifuged and free of hemolysis and particles.

Precautions】

1. Follow the instructions carefully. Results must be based on microplate reader readings.

2. If the enzyme label is not fully used, store it in a sealed bag with desiccant.

3. It is recommended to perform double testing for standards, samples, and blanks, and take the average to reduce error.

4. Remember that the sample was diluted 5 times, so multiply the result by 5 to get the actual concentration.

5. The quantitative range is 0.1–200 ng/mL. If the result is outside this range, dilute the sample with a special diluent and re-measure.

6. If the color is too light, extend the substrate incubation time slightly.

7. To prevent cross-contamination, replace pipette tips after each addition. Avoid touching the microwell plate with common reagents like the enzyme working solution, sample diluent, or substrate.

8. Do not mix reagents from different batches. Use the kit within its shelf life.

9. Substrate B is sensitive to light—keep it protected from direct sunlight.
